5-methyltetrahydrofolate (5-MTHF)

• 5-MTHF is the active metabolite of folic acid. It does not require the enzymes dihydrofolate reductase (DHFR) or methylenetetrahydrofolate reductase (MTHFR) in order to participate in reactions utilising folates as a source of one-carbon moieties.
• Folic acid is converted to dihydrofolate (DHF) by DHF reductase (DHFR) and then tetrahydrofolate (THF). Folinic acid (5-formyl THF) is rapidly metabolised via 5,10-methylenetetrahydrofolate (5,10-MTHF), which is then converted to 5-methyltetrahydrofolate (5-MTHF). See Figure 1.
• 5-MTHF can in turn be metabolised by other pathways back to 5,10-MTHF, which is converted to 5-MTHF by an irreversible enzyme MTHFR catalysed reduction using the cofactors FADH2 and NADPH.
